The woman boarded the tram outside the Adelaide train station between 4pm and 4.20pm and the man is alleged to have touched her on the leg during the voyage to Glengowrie station.
He may have been travelling with as many as three other men who also tried to photograph her with their mobile phones.
The man is described as being of Asian appearance, 165cm tall, 76kg, of slim build and with short black hair.
The woman was not injured in the incident.
Police are asking witnesses to call BankSA Crime Stoppers on 1800 333 000.
